Beschreibung: The Glioblastoma Multiforme (GBM) is the most common primary brain tumor and is currently incurable. It is urgent to devise treatments best fitting individual patients (precision medicine) and be able to predict the patients’ response to the chosen therapy. Both tumor heterogeneity and resistance to available treatments significantly affect GBM clinical management.
Our lab is countering these challenges with cellular and animal models of GBM, which are accurately reflecting genetic alterations in patients at a molecular level. In such models, we perform genetic screenings using the CRISPR/Cas9 technology as well as high-throughput drug screenings using robotics. The goal of these endeavors is the identification of molecular mechanisms regulating tumor homeostasis, which might reveal cancer-specific vulnerabilities.
Among the identified hits, we are particularly keen on prioritizing those implicated in chromatin and transcriptional regulation.
Prospective students would be involved in either validating hits from screenings or hypothesis-driven research generated by the above-mentioned experiments.
Methoden: Among the technologies to be learned and exploited:

1. Genetic engineering using CRISPR/Cas9-technology
2. Validation of potential drug target hits from in vivo screens using a variety of cell biological assays (Viability, Senescence, Fluorescent reporters) with the aid of robotic systems
3. Generation and validation of fluorescent genetic reporters

Sonstiges: Suitable candidates would ideally have a good practical background in molecular biology (cloning, DNA/RNA extraction, qPCR, Western Blotting) and/or cell biology (mammalian cell culture, cell-based assays, Flow Cytometry, Immunohistochemistry). Advanced spoken and written communication skills in English are strongly required. The minimal duration of a research internship project is 6 months.
Accepted students will get the opportunity to access and gain knowledge in state-of-the-art technologies in an international research environment. Students will be directly supervised by a staff member of the lab and the group leader.
