Beschreibung: This is a collaborative research project between the University of Hannover (Marchanka group) and the University of Frankfurt (Corzilius group), which aims to develop dynamic nuclear polarization (DNP) protocols specifically aimed at structural studies of large protein-RNA complexes by solid-state NMR.

The Centre for Biomolecular Drug Research (BMWZ) is a young and vibrant research centre (founded 2014) within the Leibniz Universität Hannover (LUH) that hosts a range of research groups in chemistry and biology. Its mission is to develop interdisciplinary approaches to basic and applied drug research by fostering collaborations between researchers both within the University and from clinical- and application-oriented partner institutions (MHH, HZI).
The Marchanka group has access to laboratory space and state-of-the-art instrumentation at the BMWZ. The NMR laboratory at the BMWZ comprises an 850-MHz Bruker AVIII HD spectrometer equipped with solution-state HCN cryo-probe and a 600-MHz NMR Bruker AVIII HD spectrometer equipped with solution-state probes and solid-state HCN MAS probes. In addition, the group has access to two further 600-MHz Bruker AVIII spectrometers equipped with solid-state HCN MAS probes. High-field and proton-detected solid-state NMR experiments are recorded at European facilities and in collaboration with national and international partners. The BMWZ also offers fully equipped laboratories for molecular biology, cell biology and biophysical measurements.

Methoden: The successful candidate for the PhD position at the University of Hannover should have Master’s degree in molecular biology, biochemistry, biotechnology, chemistry or a related field and and should have interest in biophysics and dry lab techniques (NMR and structural calculations). (S)he will express and purify proteins, will produce uniformly and specifically labelled RNA and segmentally labelled RNA, and assemble protein-RNA complexes. In addition (s)he will measure solid-state NMR spectra at the University of Hannover and DNP-enhanced MAS spectra at the University of Frankfurt, will analyse multidimensional ssNMR data (assign NMR resonances and evaluate distance restraints) and perform structure calculations.
