Beschreibung: The Schmidt lab takes an entirely novel approach to study the cell biology of the innate immune system: We generate custom-made alpaca nanobodies to interfere with and to visualize inflammasome activation in highly specific ways. Our tools render the fascinating signaling complexes of the innate immune system accessible to systematic analysis in an unprecedented way. We aim to elucidate the molecular mechanisms of inflammasome assembly and the functions of antiviral inflammasome responses against a representative panel of viruses. We address mechanistic questions from multiple perspectives using cell biological, biochemical, immunological, and virological approaches in combination with advanced imaging techniques and protein engineering.
Methoden: Biochemistry (protein purification, interaction studies)
Cell Biology (TC, inflammasome activation, flow cytometry, microscopy)
Molecular Biology (cloning, phage display)
Protein engineering (nanobodies generation)
Virology (recombinant viruses, virus production, infection experiments)
