Beschreibung: Our lab is looking for a doctoral student in the field of computational structural biology and protein evolution. Understanding binding energetics of protein-protein interactions at the molecular and structural level is not only interesting for basic science but also valuable for biotechnological applications such as drug design. Early studies showed that different binding interface residues have various importance to binding energetics and only a few key residues, referred to as binding hot-spots, are the main contributors to the binding free energy. In contrast to binding hot-spots, we recently proposed a new notion of binding “cold-spots” or positions in the protein-protein interface that are occupied by non-optimal amino acids and could bring substantial improvement in affinity upon mutation. Identification of such positions is crucial for studies of protein evolution and for protein design. The particular project aims at analyzing multiple structures of protein-protein interactions, searching for cold-spots in these interactions and analyzing various structural scenarios for cold spot occurrence. We are looking for a motivated person with understanding of structural biology, experience in programming and familiarity with various computational structural biology tools.
Methoden: Computational structural biology, analysis of protein structures, development of computational methods for analysis and design of protein-protein interactions.
