Beschreibung: In neurons, mRNA localization and local translation at pre- and postsynaptic areas has been shown to regulate synaptic plasticity and learning. For transport, mRNAs are recognized by dedicated RNA-binding proteins, which themselves are tethered to cytoskeletal motor proteins. Two human core RNA-binding proteins of such transport particles are Staufen2 and Pumilio2. The aim of this PhD thesis is to dissect the molecular binding modes of the RNA-binding proteins by using structural biology, biochemistry, and cell biology.

- Appropriate studies (Master/Diploma) in biochemistry, biology, biotechnology or related disciplines
- Expertise in biochemistry
- Interest in structural biology
- Substantial knowledge in molecular biology
- Expertise in cell culture is of advantage
- Strong motivation, flexibility, and team work
Methoden: - Recombinant expression and purification of Staufen2 and Pumilio2.
- Large-scale in vitro transcription of target RNAs and quantitative interaction studies with proteins.
- Crystallization screens and structure determination of Staufen2 and/or Pumilio2 with target RNAs using X-ray crystallography and micro-electron diffraction.
- Quantitative interaction studies of proteins with structure-guided mutations using biophysical approaches (e.g. ITC, SPR).
- Cell-culture based identification of protein interactors of Staufen2 and Pumilio2 using BioID.
