Beschreibung: Our group has identified and characterized a sortase from the opportunistic pathogen Propionibacterium acnes (10.1042/BCJ20180885). Sortases are highly relevant biotechnological enzymes as they allow to specifically label proteins, e.g. for the production of antibody drug conjugates. In contrast to most sortase enzymes, the sortase F from P. acnes is highly active. Nevertheless, improved variants of the sortase F enzyme are needed for the commercial application of the enzyme.

In the master thesis project the student will build up a yeast based screening assay for sortase transpeptidation activity. This assay will then allow to evolve highly active variants of sortase F. The project involves a broad spectrum of state-of the-art techniques including molecular biology work, cloning, assay development and protein purification.

The University of Applied Sciences Northwestern Switzerland in Muttenz (Canton Basel-Landschaft) offers modern and well-equipped laboratories and is situated in one of the most prominent centres of the Life Sciences Industry. We are located in the vivid trinational region (Switzerland, Germany, and France) in a beautiful landscape.
