Beschreibung: Heart failure is the leading cause of death worldwide. The limited regenerative capacity of the human heart is a major factor in the high morbidity and mortality rates. In contrast, the zebrafish exhibits an ability to regenerate its heart and, thus, can provide critical insights as to how regeneration is achieved or might be stimulated. Chromatin regulation is fundamental to cell proliferation, fate determination and differentiation during embryonic development and in mediating cellular responses to the environment or upon injury. Although considerable knowledge exists about the transcriptional changes accompanying zebrafish heart regeneration, little is known how chromatin states and gene expression patterns are coordinated during the regenerative process. Using genome wide ChIP-Seq and RNA-Seq approaches we defined the dynamic epigenetic and transcriptomic landscapes during zebrafish heart regeneration and investigated how global patterns of gene expression and chromatin organization are coordinated. We uncovered novel cis-regulatory elements that integrate transcriptional inputs deployed during cardiac regeneration. The aim of the project is to dissect the function of enhancers and upstream transcription factors that we identified in our study in cardiac regeneration.
Methoden: We use a wide range of biochemical, molecular/cell biological, embryological and transgenic techniques to study cardiac development and regeneration in mouse and zebrafish.
