Beschreibung: G protein-coupled receptors (GPCRs) have an outstanding role in pharmaceutical research and are the target molecule of about 40% of the currently available drugs. In their function for signal transduction, a ligand usually binds on the extracellular side of the receptor and initiates conformational changes within the seven transmembrane helices of GPCRs which leads to the binding of G proteins on the intracellular side. Signaling on the G protein pathway can be blocked by intracellular binding arrestins that cause internalization of the receptor via endocytosis. While the structure-function relationship of G protein-GPCR-complexes are describe quite well, structural data of the complex of arrestin and GPCRs, especially peptide-binding GPCRs, are sparse.
The aim of the proposed research project is to describe key interactions between arrestin and the Y receptors and the orientation of these two proteins to each other. For this, solution and solid-state NMR as well as EPR spectroscopy will be employed. Furthermore, changes of the dynamic properties of the receptors after formation of the complex with ligand and arrestin will be characterized. This will add up to a deeper understanding of the function of the Y receptors.
Methoden: We are looking for a motivated and open-minded candidate with the ability to work independently as well as in a team, with a strong interest in GPCR biology and biophysical methods. A very good level of written and spoken English is mandatory. Methodological, the project comprises recombinant protein expression, protein purification, reconstitution of receptors into lipid bicelles as well as NMR and EPR spectroscopy. Expertise with these methods is no prerequisite, but a benefit.
