Beschreibung: Mitochondria are important organelles that are essential for energy conversion and many other important cellular functions. Mitochondrial structure is highly versatile and undergoes dynamic adaptations upon metabolic or bioenergetic cues. We recently show that cristae membrane undergoes continuous cycles of dynamic remodelling in a timescale of seconds that is dependent on a large oligomeric complex called MICOS. MICOS complex regulates formation of crista junctions and contacts between inner and outer mitochondrial membrane. Several subunits of MICOS complex have been identified and mutations in some of them are associated with human diseases like Parkinson’s disease, mitochondrial encephalopathy with liver failure and mitochondrial myopathies. We want to address the fundamental question of how cristae and crista junctions are formed by MICOS subunits and how do they influence the progression of human diseases.
Methoden: We use a combination of following techniques: mammalian cell culture, CRISPR-Cas methods for generation of knockout cells, biochemical assays, molecular biology tools, electron microscopy, fluorescence imaging including super-resolution nanoscopy and analysis methods.

A DFG-funded PhD position (E13, 65%) for a period of three years is available.
