Beschreibung: We are looking for applicants for a PhD-position in the group of Prof. Andreas Meisel and Dr. Siegfried Kohler at the Department of Experimental Neurology at the Charité Universitätsmedizin Berlin, Germany. The work for this project is in close collaboration with the group of Prof. Falk Hiepe (Department of Rheumatology and Clinical Immunology at Charité & German Rheumatism Research Center, DRFZ).
The project focuses at exploring new therapeutic strategies for Myasthenia Gravis and other autoantibody-mediated diseases. Current therapies are not curative and have the disadvantages of serious side effects and increased risk of infection due to suppression of protective immunity. To overcome this limitation, we have developed special antibody-protein conjugates to deplete autoantibody producing plasma cells selectively in vivo (Cheng, Pelz, et al.; Eur J Immunol. 2020 Feb;50(2):284-291. doi: 10.1002/eji.201948144.). The next goal is the transfer of this technology to an autoimmune disease for the therapeutic depletion of antigen specific plasma cells in the experimental autoimmune Myasthenia Gravis (EAMG) model. Additional experiments with samples from Myasthenia Gravis patients will lay the foundation for a new therapy for human Myasthenia Gravis and other autoantibody-mediated diseases.
Methoden: Your responsibilities:
- Induction of the experimental autoimmune myasthenia gravis (EAMG) mouse model, assessment of disease state and treatment of mice (mouse handling, injections, blood collection)
- Generation of antibody-protein conjugates and their characterization (molecular cloning, SDS-PAGE and Western Blot, ELISA)
- Analysis of in vitro and in vivo plasma cell depletion experiments (cell culture, flow cytometry, ELISpot)
- Data collection, analysis and interpretation (MS Office suite, statistical programs)

- M.Sc. or equivalent in biology, biochemistry, biomedicine or a similar field
- Experience with animal experiments is desired (FELASA B certificate would be a plus)
- First experiences in some of the above mentioned methods
- Highly motivated, self-organized and reliable
- Solid background and interest in immunology and autoimmunity
This work is part of third-party funding projects and financed for a period for 3 years.
