Beschreibung: We are recruiting a candidate to develop a new strategy to treat the brain tumor glioblastoma that includes research with novel chemical protein modifications and molecular biology. The Berlin Universities Alliance (BUA) in collaboration with the University of Melbourne (UoM) is offering unique and exciting opportunities for two potential PhD candidates, one with their home at Charité – Universitätsmedizin Berlin (BUA) and the other at UoM. The overall research program investigates the site-specific coupling via glycans in Berlin and demonstration of efficacy in animal models in Melbourne. The Charité based PhD study will focus on artificial glycosylation of proteins in vitro, toxin expression in bacteria and coupling of the toxin to glycosylated nanobodies. Each candidate will spend a minimum of 12 months at the partner institution mid-term. The PhD candidates will be jointly supervised.
Methoden: Responsibilities

Unique targeted toxins, based on an antibody and nanobody against calcitonin receptor developed at UoM, will be synthesized and refined by the group at Charité, with expertise in click chemistry, plant toxins and endosomal escape. The PhD candidate at Charité will focus on artificial glycosylation of proteins in vitro, toxin expression in bacteria and coupling of the toxin to glycosylated nanobodies. Novelty includes a new site-specific coupling technique that allows the generation of homogenous tailormade products in contrast to current antibody-drug conjugates that are heterogenous and undefined in structure. The position will be remunerated in the form of a tax-free stipend of €1500 per month and will include up to €7000 for the travel to and stay in Melbourne. Your responsibilities are:

• Development of a method for site-specific modification of a nanobody with a glycan containing a click-chemistry group
• Expression of a protein toxin in bacterial expression systems
• Site-specific modification of the toxin with a click-chemistry adapter and production of an immunotoxin by click-chemistry
• Analytical and functional studies on the product and independent evaluation and documentation of results
• Publication of results in scientific journals and attendance of scientific conferences and presentation of results
• Supervision of bachelor and/or master theses in the same project
• Constant exchange with the partner project in Melbourne and joint experimental design
• Obligatory research stay at the partner university in Melbourne for at least 12 months

Profile

• A Master in molecular biology or biochemistry with strong chemical background (click-chemistry) or a Master in (click-) chemistry with strong biochemical background (protein expression and modification)
• The candidates will be enrolled in the Institut für Laboratoriumsmedizin, Klinische Chemie und Pathobiochemie at Charité – Universitätsmedizin Berlin and in the Department of Medicine – Austin Health PhD program at the University of Melbourne, and must therefore fulfill the requirements of both institutions
• Demonstrated experience in the field of click-chemistry, glycan chemistry, amino acids, bacterial protein expression, and preferably molecular biology
• Demonstrated knowledge of analytical methods (HPLC, MALDI, NMR, TLC, electrophoresis, Westernblot, etc.)
• Demonstrated ability to work independently and as part of a multidisciplinary and international team, open-mindedness
• Demonstrated time, management, and written and oral communication skills
• Demonstrated ability to write research reports or other publications in a scientific standard
• Profound knowledge of written and spoken English, basic knowledge of German or the willingness to learn it
• Willingness and motivation for a research stay in Melbourne of at least one year
• Compliance with German and Australian guidelines related to Covid-19 (mandatory vaccination, etc.) and general visa requirements
