Our immune system incessantly defends us against infections. T-cells recognize foreign antigens on the surface of antigen-presenting cells (APCs) and activate to initiate an immune response. The molecular details of this process, however, remain unclear due to the complex and dynamic interface between T-cells and APCs.
In this project, we will recreate T-cell antigen recognition by interfacing live T-cells with synthetic APCs. These sAPCs enable us to simplify the complex interaction and precisely control APC properties, including the organization of antigens and other involved proteins, and physical characteristics like softness.
The task of the successful candidate is to design and produce microparticles, which are the basis for sAPCs. The material platform will have to offer the possibility for a photo-orthogonal de-/crosslinking of the hydrogel to adjust the compliance of the sAPCs during LLSM experiments.
The project is being carried out in close cooperation with biophysicists from the Institute of Applied Physics of TU Wien.
